ABSTRACT

The aim of this study was to construct Chlorella mutants deficient in chlorophyll synthesis by atmospheric pressure room temperature plasma (ARTP) mutagenesis, and screen novel algal species with very low chlorophyll content which is suitable for protein production by fermentation. Firstly, the lethal rate curve of mixotrophic wild type cells was established by optimizing the mutagenesis treatment time. The mixotrophic cells in early exponential phase were treated by the condition of over 95% lethal rate, and 4 mutants with the visual change of colony color were isolated. Subsequently, the mutants were cultured in shaking flasks heterotrophically for evaluation of their protein production performance. P. ks 4 mutant showed the best performance in Basal medium containing 30 g/L glucose and 5 g/L NaNO3. The protein content and productivity reached 39.25% dry weight and 1.15 g/(L·d), with an amino acid score of 101.34. The chlorophyll a content decreased 98.78%, whereas chlorophyll b was not detected, and 0.62 mg/g of lutein content made the algal biomass appear golden yellow. This work provides a novel germplasm, the mutant P. ks 4 with high yield and high quality, for alternative protein production by microalgal fermentation.

ABSTRACT

Phytoremediation plays an important role in the treatment of heavy metal pollution in soil. In order to elucidate the mechanism of salicylic acid (SA) on copper absorption, seedlings from Xuzhou (with strong Cu-tolerance) and Weifang Helianthus tuberosus cultivars (with weak Cu-tolerance) were selected for pot culture experiments. 1 mmol/L SA was sprayed upon 300 mg/kg soil copper stress, and the photosynthesis, leaf antioxidant system, several essential mineral nutrients and the changes of root upon copper stress were analyzed to explore the mechanism of copper resistance. The results showed that Pn, Tr, Gs and Ci upon copper stress decreased significantly compared to the control group. Meanwhile, chlorophyll a, chlorophyll b and carotenoid decreased with significant increase in initial fluorescence (F0), maximum photochemical quantum yield of PSⅡ (Fv/Fm), electron transfer rate (ETR) and photochemical quenching coefficient (qP) content all decreased. The ascorbic acid (AsA) content was decreased, the glutathione (GSH) value was increased, the superoxide dismutase (SOD), catalase (CAT) and ascorbate peroxidase (APX) activity in the leaves were decreased, and the peroxidase (POD) activity was significantly increased. SA increased the Cu content in the ground and root system, and weakened the nutrient uptake capacity of K, Ca, Mg, and Zn in the root stem and leaves. Spray of exogenous SA can maintain the opening of leaf stomata, improve the adverse effect of copper on photosynthetic pigment and PSⅡ reaction center. Mediating the SOD and APX activity started the AsA-GSH cycle process, effectively regulated the antioxidant enzyme system in chrysanthemum taro, significantly reduced the copper content of all parts of the plant, and improved the ion exchange capacity in the body. External SA increased the content of the negative electric group on the root by changing the proportion of components in the root, promoted the absorption of mineral nutrient elements and the accumulation of osmoregulatory substances, strengthened the fixation effect of the root on metal copper, and avoided its massive accumulation in the H. tuberosus body, so as to alleviate the inhibitory effect of copper on plant growth. The study revealed the physiological regulation of SA upon copper stress, and provided a theoretical basis for planting H. tuberosus to repair soil copper pollution.

ABSTRACT

The present study reports on seasonal and spatial variations in diversity, distribution and abundance of dinoflegellates and indicates the presence of HAB species in Pakistan waters. A total of 179 taxa, recorded in this study from offshore and near-shore waters, belong to 41 genera in 26 families and 10 orders. The high species count (149 species) was recorded from Manora Island offshore station (MI-1) and 105 spp, 109 spp and 115 spp were encountered from the Mubarak village offshore station (MV-1), Manora near shore station (MI-2) and Mubarak Village near-shore station (MV-2) respectively. Tripos furca was the dominant and frequently occurring species (> 1 x103 to > 25 x103 cells L-1 from coastal and >1x 105 cells L-l from near-shore stations) in addition to less abundant Alexandrium catenella, Alexandrium sp., Alexandrium minutum, and Prorocentrum micans (>103 to 25x 103cells/L). Another 44 species occurred in relatively low numbers (<103 cell L-l). Seventy species were found throughout the study period at all four stations. High number of species in three genera (Tripos (38), Protoperidinium (34) and Prorocentrum (20) was recorded. Potently toxic (16 genera 43 species) and HAB related (19 genera and 30 species) dinoflagellate taxa were also recorded. The percent contribution of dinoflagellates in total phytoplankton population generally remained below 20% except for a few instances. Manora Island stations had comparatively higher Shannon index and equitability and slightly lower dominance index. The PCA plot showed strong positive correlation among chlorophyll-a concentration, dissolved oxygen, total number of phytoplankton and dinoflagellates.


O presente estudo relata variações sazonais e espaciais na diversidade, na distribuição e na abundância de dinoflegelados e indica a presença de espécies de HAB nas águas do Paquistão. Um total de 179 táxons, registrados nesse estudo de águas offshore e próximas à costa, pertence a 41 gêneros em 26 famílias e 10 ordens. A alta contagem de espécies (149 espécies) foi registrada na estação offshore da Ilha de Manora (MI-1) e 105 spp., 109 spp. e 115 spp. foram encontrados na estação offshore da vila de Mubarak (MV-1), Manora perto da estação costeira (MI- 2) e estação próxima à costa da Vila de Mubarak (MV-2), respectivamente. Tripos furca foi a espécie dominante e de ocorrência frequente (> 1 x103 a > 25 x103 células L-1 da costa e > 1x 105 células Ll de estações próximas à costa), além de Alexandrium catenella menos abundante, Alexandrium sp., Alexandrium minutum e Prorocentrum micans (> 103 a 25x 103 células/L). Outras 44 espécies ocorreram em números relativamente baixos (< 103 células L-1). Setenta espécies foram encontradas durante o período de estudo em todas as quatro estações. Foi registrado um alto número de espécies em três gêneros (Tripos (38), Protoperidinium (34) e Prorocentrum (20). Potencialmente tóxicos (16 gêneros e 43 espécies) e HAB relacionados (19 gêneros e 30 espécies), táxons de dinoflagelados também foram registrados. A contribuição percentual de dinoflagelados na população fitoplanctônica total geralmente permaneceu abaixo de 20%, exceto em alguns casos. As estações da Ilha de Manora tinham índice de Shannon comparativamente mais alto e equitabilidade e índice de dominância ligeiramente mais baixos. O gráfico de PCA mostrou forte correlação positiva entre concentração de clorofila-a e oxigênio dissolvido, número total de fitoplânctons e dinoflagelados.

ABSTRACT

Abstract Silicon (Si) is an element that can improve the growth and development of rice plants in water-deficient environments because it is an enzymatic stimulant, signaling for production of antioxidant compounds. Therefore, the aim of this study was to examine the relationship between water deficiency and the effect of Si on two rice cultivars whose seeds were treated with dietholate. The experimental design was fully randomized with three replicates, and treatments were organized in a 3x2x2x4 factorial arrangement: three water soil conditions (50% and 100% of soil water retention capacity (WRC) and complete submergence in a water blade of 5.0 cm); two cultivars (IRGA 424 RI and Guri INTA CL); two sources of Si (sodium metasilicate and potassium metasilicate); and four rates of Si (0; 4.0; 8.0 and 16 g L-1). Chlorophyll a and b, leaf area and shoot and root dry weight increased at higher rates of Si under the three soil water regimes. There was an increase in superoxide dismutase and guaiacol peroxidase enzyme activity in the cultivars at higher rates of Si, reducing lipid peroxidation caused by water deficiency. Therefore, Si did indeed attenuate water deficiency stress in rice plants emerging from seeds treated with dietholate.


Resumo O silício (Si) é um elemento que pode proporcionar melhor crescimento e desenvolvimento às plantas de arroz cultivadas em ambientes com deficiência hídrica, por ser um estimulador enzimático, promovendo sinalização para produção de compostos antioxidantes. Assim, o objetivo deste trabalho foi estudar a relação entre deficiência hídrica e o efeito do Si em duas cultivares de arroz tratadas com dietholate. O delineamento experimental utilizado foi inteiramente casualizado, com três repetições e os tratamentos arranjados em esquema fatorial 3x2x2x4, sendo os fatores: três condições hídricas do solo (50%, 100% da capacidade de retenção de água no solo (CRA) e lâmina d'água de 5,0 cm), duas cultivares (IRGA 424 RI e Guri INTA CL), duas fontes de Si (metassilicato de sódio e metassilicato de potássio) e quatro doses de Si (0; 4,0; 8,0 e 16 g L-1). O índice de clorofila a e b, a área foliar e o teor de massa seca da parte aérea e raiz aumentaram com o aumento das doses de Si nas três condições hídricas do solo. Houve um aumento na atividade das enzimas superóxido dismutase e guaiacol peroxidase nas cultivares estudadas à medida que as doses de Si aumentaram, diminuindo a peroxidação de lipídios, causada pela restrição hídrica. Portanto, o Si atenua o estresse por déficit hídrico em plantas de arroz emergentes de sementes tratadas com dietholate.

ABSTRACT

The flower of Polygonatum cyrtonema has good edible and medicinal values. In this study, four samples of P. cyrtonema flowers from different regions were selected as test materials. The contents, composition and antioxidant activities of lipid-soluble pigments and alcohol-soluble components were determined under different light and temperature conditions, which help to reveal the discoloration reason and the composition variation patterns during storage. The results showed that light and temperature had different effects on the lipid-soluble pigments and alcohol-soluble components in the dried flowers during storage. After storage for 4 weeks, the contents of total chlorophyll, carotenoids, phenols and saponins in the samples exposed to light respectively decreased by 62.62%, 66.4%, 68.7% and 43.4% compared with those in the dark. The decreases in the contents of chlorophyll a, chlorophyll b, lutein, β-carotene and zeaxanthin were 64.64%, 56.74%, 59.2%, 77.7% and 45.4%, respectively. The contents of pigments and components in the samples stored at-20 ℃ were significantly higher than those at room temperature and 4 ℃, indicating that low temperature was conductive to the stability of lipid-soluble pigments and alcohol-soluble components. The samples stored at low temperature and in the dark had the strongest free radical scavenging activity. The results suggest that P. cyrtonema dried flowers should be stored in low temperature environment without light, which can slow down the degradation of internal components. The study provides a theoretical basis for the production, processing and storage of P. cyrtonema flowers.

The aluminum in high levels in the soil affects the emergence, growth, and development of various species. Therefore, the objective of this study was to evaluate the emergence and physiological behavior of four provenances of Jatropha curcas subjected to different levels of aluminum. The experiment was performed in a greenhouse in a completely randomized design, with four levels of aluminum in the soil (8.2, 16.5, 24.0 mmolc·dm-3 and control) and four provenances of J. curcas seeds (P1 = Dourados-MS, P2 = Montes Claros-MG, P3 = Alta Floresta-MT, and P4 = Petrolina-PE); the effects of aluminum toxicity were investigated in 25, 50, 75, and 100 days after emergence. The levels of aluminum in the soil were collected from the initial soil correction, which featured an aluminum level of 24.0 mmolc dm-3. The seedling emergence was not affected by treatment with aluminum; however, the height and leaf area of P1, P2, and P3 were reduced with increasing levels of aluminum. The emergence and vigor of J. curcas seeds were not influenced by the differences in the origins of the seeds or by the aluminum levels evaluated. Gas exchanges were affected negatively by aluminum and the responses of the chlorophyll a fluorescence indicate harmful effect in the photosynthetic apparatus. The seeds of origin P4 (Petrolina-PE) has increased tolerance to stress conditions.


O alumínio em níveis elevados no solo afeta a emergência, o crescimento e o desenvolvimento de diversas espécies. Diante disso, o objetivo desse trabalho foi avaliar a emergência e o comportamento fisiológico de quatro procedências de Jatropha curcas submetidas a diferentes níveis de alumínio. O experimento foi conduzido em casa de vegetação, em delineamento inteiramente casualizado, com quatro níveis de alumínio no solo (8,2; 16,5 e 24,0 mmolc dm-3 e o controle) e quatro procedências de sementes de J. curcas (P1 = Dourados-MS P2 = Montes Claros-MG, P3 = Alta Floresta-MT e P4 = Petrolina-PE), e os efeitos da toxicidade do alumínio foram investigados aos 25, 50, 75 e 100 dias após a emergência. A emergência das plântulas não foi afetada pelo tratamento com alumínio; no entanto, a altura e a área foliar de P1, P2 e P3 foram reduzidas com níveis crescentes de alumínio. A emergência e o vigor das plântulas das procedências não foram afetados pelos níveis de alumínio avaliados. A taxa fotossintética, taxa de transpiração e condutância estomática também foram reduzidas quando as plantas foram cultivadas em solo contendo altas níveis de alumínio. As plantas P4 foram menos sensíveis aos níveis crescentes de alumínio.

ABSTRACT

Unmanned aerial vehicle(UAV) remote sensing and vegetation index have great potential in the field of Chinese herbal medicine planting. In this study, the visible light image of Polygonatum odoratum planting area in Changyi district of Jilin province were acquired by UAV, and the real-time monitoring of P. odoratum planting area was realized. The green leaf index(GLI) was established, and GLI values of P. odoratum were collected used the spatial sampling points. To compare the GLI values in different periods, it was found that the GLI values of P. odoratum have three stages changing rule of rising-gentle-falling related to the germination, vigorous growth and withered of P. odoratum growth. Meanwhile, the GLI values were compared with four biomass data of P. odoratum, including plant height, leaf area, chlorophyll a and chlorophyll b content in leaves, and it was found that the GLI value was related to the growth potential of P. odoratum. The GLI value with a rapid increase in rising stage or at a high level in the gentle stage means the P. odoratum was in a better growth potential. GLI value has a same change trend with plant height, and has certain correlation with plant height and leaf area. However, there is no obvious relationship between chlorophyll a and chlorophyll b contents in leaves and GLI value. The study clarified the change rule of GLI value of P. odoratum, explained the reason for the change of GLI value, and expanded the application range of GLI. The research shows that UAV and vegetation index can be applied to monitoring the Chinese herbal medicines planting, and provides a new idea for exploring more effective information extraction methods of Chinese herbal medicines.

ABSTRACT

The purpose of this experiment was to study the effects of different shading conditions on the growth,physiological characteristics and biomass allocation of Polygonatum cyrtonema,which offered a theoretical basis for its cultivation.Different light environments(100%,80%,60% and 35% light transmittance) were simulated with shading treatments.Growth and photosynthetic indexes of P.cyrtonema were measured and the variances were analyzed.The results show that shading decreased superoxide anion radical(O-·2)production rate and hydrogen peroxide(H_2O_2) accumulation,kept the activity of SOD,POD and CAT enzyme at a high level.Furthermore,The content of chlorophyll a and chlorophyll b,net photosynthetic rate(Pn),stomatal conductance(Gs),transpiration rate(Tr),maximal photochemical efficiency of photosystem Ⅱ(Fv/Fm),photochemical quenching index(q P) and effective quantum yield of photosystem II(ΦPSⅡ) of P.cyrtonema were increased while the intercellular CO2 concentration(Ci),Foand NPQ were decreased by shading.Shading is beneficial to P.cyrtonema growth,can increase the total biomass P.cyrtonema.The allocation proportion of biomass on the aerial portion of P.cyrtonema increased but underground parts decreased with increasing shading conditions.In this study,P.cyrtonema can grow well in shading conditions,shading is beneficial to the formation of the yield and quality of the rhizomes of P.cyrtonema,especially in 65% light transmittance.

ABSTRACT

Abstract Gall-inducing insects manipulate the structural, histochemical and physiological profiles of host-plant tissues to develop galls. We evaluated galls induced by Eugeniamyia dispar on the leaves of Eugenia uniflora in an attempt to answer the following questions: (i) How does this gall-inducing insect change the structural and histochemical profiles of the host-plant organ? (ii) Despite structural changes, can gall tissues maintain photosynthetic activity? Starch, proteins, reducing sugars and reactive oxygen species were detected mainly in the nutritive tissue surrounding the larval chamber. Despite structural changes, the galls induced by E. dispar on E. uniflora retain chlorophyllous tissue, although its amount and photosynthetic activity are less than that of non-galled leaves. This reduced photosynthetic activity, in association with the presence of large intercellular spaces, could improve gas diffusion and, consequently, avoid hypoxia and hypercarbia in gall tissue.(AU)


Resumen Los insectos que inducen las agallas manipulan los perfiles estructurales, histoquímicos y fisiológicos de los tejidos de la planta hospedera para su desarrollo. Nosotros evaluamos las agallas inducidas por Eugeniamyia dispar en las hojas de Eugenia uniflora en un intento de responder las siguientes preguntas: (i) ¿Cómo este insecto inductor de agallas cambia los perfiles estructurales e histoquímicos en el órgano de la planta hospedera? (ii) A pesar de las modificaciones estructurales, ¿pueden los tejidos de la agalla mantener la actividad fotosintética? El almidón, las proteínas, los azúcares reductores y las especies reactivas de oxígeno se detectaron principalmente en la capa de tejido nutritivo que rodea a la cavidad larval. A pesar de las modificaciones estructurales, las agallas inducidas por E. dispar en E. uniflora retienen su tejido clorofílico, aunque su cantidad y actividad fotosintética son menores que en las hojas no agalladas. Esta actividad fotosintética reducida, asociado a la presencia de grandes espacios intercelulares, pueden mejorar la difusión de gases y, en consecuencia, evitar la hipoxia y la hipercapnia en los tejidos de las agallas.(AU)

Abstract Current study investigated the effectiveness of different macrophytes as culture media for Ankistrodesmus gracilis in laboratory conditions. Significant difference (p < 0.05) was reported in cell density with regard to conventional culture medium (CHU12) and macrophytes culture media. Mean cell density in NPK, Eichhornia crassipes and E. azurea media was higher (p < 0.05) than in conventional culture medium. Chlorophyll-a was higher than 1 g.L-1, except in CHU12 (0.7 ± 0.4 g.L-1) and T. domingensis (0.8 ± 0.3 g.L-1) media. Nitrate decreased sharply as from the 7th-day of the experiment. Ammonium and total phosphorus were highest in culture media and ranged between 0.4 g.L-1 (P. cordata medium) and 1.7 g.L-1 (CHU12 medium) for ammonium, and between 0.8 g.L-1 (CHU12 medium) and 1.9 g.L-1 (T. domingensis medium) for total phosphorus. Results revealed inorganic fertilizer and macrophytes combined with vitamins may be effective as culture media and strongly supports the growth of Ankistrodesmus gracilis, since cell density and biochemical composition are similar to or higher than conventional culture medium (CHU12). Macrophyte is a tool for aquaculture since biological wastes may be used with nutrients to improve the cultivation of microalgae.


Resumo O presente estudo investigou a eficácia na utilização de diferentes macrófitas como meio de cultura para Ankistrodesmus gracilis em condições laboratoriais. Foi observada diferença significativa (p < 0,05) entre a densidade celular em relação aos meios de cultura convencional (CHU12) e de macrófitas. Os meios de cultura com Eichhornia crassipes, E. azurea e NPK apresentaram densidade celular maiores (p < 0,05) que o meio de cultura convencional. Os teores de clorofila-a foram superiores a 1 g.L-1, exceto nos meios de cultura CHU12 (0.7 ± 0.4 g.L-1) e de T. domingensis (0.7 ± 0.3 g.L-1). O nitrato diminuiu acentuadamente a partir do 7º dia de experimento. Dentre os nutrientes, amônia e o fósforo total foram os mais elevados nos meios de cultura, variando entre 0.4 g.L-1 (meio de P. cordata) a 1,7 g.L-1 (meio CHU12) para amônia e, entre 0.8 g.L-1 (meio CHU12) e 1,9 gL-1 (meio de T. domingensis) para o fósforo total. Os resultados do presente estudo mostraram que o uso de fertilizantes inorgânicos e macrófitas, combinados com vitaminas, pode ser eficaz como meio de cultura no crescimento de Ankistrodesmus gracilis, uma vez que a densidade celular e a composição bioquímica foram semelhantes ou superiores ao meio de cultura convencional (CHU12). As macrófitas são ferramentas a serem adotadas na aquicultura, desde que os resíduos biológicos podem ser usados como nutrientes para melhorar o cultivo de microalgas.

ABSTRACT

Abstract The Llanquihue lake is included in the called Araucanian or Nord Patagonian lakes located between 38-41° S. These lakes are characterized by their oligo-mesotrophic status due to human intervention which takes to the increase in nutrients inputs from industries and towns. Effects on zooplankton assemblages are observed with marked increase of daphnids abundance. The aim of the present study is to analyze the trophic status and zooplankton relative abundance in different bays of Llanquihue lake. It was found direct associations between chlorophyll a with daphnids percentage, total dissolved nitrogen with reactive soluble phosphorus nitrogen/phosphorus molar radio with cyclopoids percentage, and an inverse relation between daphnids and calanoids percentages. The occurrence of three kinds of microcrustacean assemblages and environmental conditions was evidenced: the first one with high calanoids percentage, low species number and low chlorophyll and nutrients concentration, a second with moderate chlorophyll and nutrients concentration and moderate daphnids percentage; high species number and a third site with high chlorophyll and nutrients concentration, high daphnids percentage and high species number. Daphnids increase under mesotrophic status, agree with similar results observed for southern Argentinean and New Zealand lakes.


Resumo O lago Llanquihue está incluído nos chamados lagos araucana ou Nord Patagônia localizado entre 38-41° S. Estes lagos são caracterizados pela condicao oligo-mesotrofica debido a intervencao humana, com aumento da carga de nutrientes provenientes de industrias y areas urbanas com efeitos sobre as assembleias zooplantonicas sao observadas, com aumento acentuado de dafnideos. O objetivo do presente estudo é analisar o estado trófico a abundancia relative do zooplancton em diferentes compartimentos do lago Llanquihue. Foram encontradas associações diretas entre clorofila a com percentual de dafinídeos, nitrogênio total dissolvido com fósforo solúvel reativo molares razao molar nitrogênio / fósforo com percentual de ciclopóides, e uma relação inversa entre percentuais de dafinídeos e calanóides porcentagens. A ocorrência de três tipos de assembleias de microcrustáceos e as condições ambientais fora: a primeira com alta porcentais de calanóides, baixo número de espécies e baixa clorofila e a nutrientes, uma segunda com concentracoes moderadas de clorofila e nutrientes percentual moderado de daphnideos e alto número de espécies; e uma terceiro local com alta concentração de clorofila e nutrientes, alta abundância dafinídeos e número elevado de espécies. Resultados similares com aumento de dafnideos em condicoes mesotroficas também foram observados para lagos da Argentina e Nova Zelândia do sul.

ABSTRACT We studied the effects of particulate and dissolved optically active components on the attenuation of photosynthetic active radiation (PAR) in a tropical lake. The temporal and spatial distribution of tripton, Chl-a and aCDOM(440) and their relative contribution to the diffuse PAR attenuation coefficient (Kd) was investigated at 21 sites (dry and wet seasons and two intermediate periods) and at monthly interval at 1 pelagic site. Higher values of ​​ Kd were observed during the mixing period, characterized by a higher concentration of tripton and Chl-a compared to the stratified rainy season. In the spatial sampling PAR attenuation was dominated by tripton absorption/scattering (average relative contribution of 79%), followed by Chl-a (average 11.6%). In the monthly sampling tripton and Chl-a accounted for most of the Kd with relative contributions of 47.8% and 35.6%, respectively. Multiple linear regression analysis showed that Chl-a and tripton in combination explained 97% of the monthly variation in Kd (p<0.001), but Chl-a had more influence (higher regression coefficient). Thus, although most of light attenuation was due to tripton, seasonal variations in phytoplankton abundance were responsible for most of the temporal fluctuations in Kd.

ABSTRACT

Abstract This study aims to evaluate the environmental factors determining of the changes in phytoplankton structure in spatial (upper, middle and lower course) and seasonal (dry and rainy period) scales in a eutrophic river (Almada River, northeastern Brazil). In the study period, total accumulated rainfall was below of the historic average, resulting in flow reduction, mainly in rainy period. High orthophosphate concentration was found at the sampling sites. Phytoplankton chlorophyll a increased from upstream to downstream. Geitlerinema splendidum (S1) and Chlamydomonas sp. (X2) were the most abundant species in the upper course and several species of diatoms (D), Euglenophyceae (W1, W2) and Chlorophyceae (X1) in the middle and lower course. The functional groups were found to be characteristic of lotic ecosystem, shallow, with low light availability, rich in organic matter and eutrophic environments. We conclude that phytoplankton community structure was sensitive to change of the river flow and nutrient availability in spatial and seasonal scale in a tropical river.


Resumo O objetivo deste estudo foi avaliar os fatores ambientais determinantes das mudanças na estrutura do fitoplâncton em escala espacial (alto, médio e baixo curso) e temporal (seca e chuva) em um rio eutrófico tropical (rio Almada, nordeste do Brasil). No período de estudo, a precipitação acumulada mensal foi abaixo da média histórica, resultando na redução da vazão, principalmente no período chuvoso. A concentração de ortofosfato foi elevada nos pontos de amostragem. A clorofila a do fitoplâncton aumentou de montante para jusante. Geitlerinema splendidum (S1) e Chlamydomomas sp. (X2) foram as espécies mais abundantes no alto curso e várias espécies de diatomáceas (D), Euglenophyceae (W1, W2) e Chlorophyceae (X1) no médio e/ou baixo curso. Os grupos funcionais foram característicos de ambientes lóticos, rasos, com baixa disponibilidade de luz, rico em matéria orgânica e eutrófico. Concluímos que a estrutura da comunidade fitoplanctônica foi sensível às mudanças na vazão e na disponibilidade de nutrientes em escala espacial e temporal em um rio tropical.

Abstract Satellite images are an effective tool for the detection of phytoplankton blooms, since they cause striking changes in water color. Bloom intensity can be expressed in terms of chlorophyll-a concentration. Previous studies suggest the use of Landsat TM4/TM3 reflectance ratio to retrieve surface chlorophyll-a concentration from aquatic systems. In this study we assumed that a remote sensing trophic state index can be applied to investigate how changes in HRT along the hydrologic year affect the spatial distribution of the phytoplankton blooms at Ibitinga’s reservoir surface. For that, we formulated two objectives: (1) apply a semi-empirical model which uses this reflectance ratio to map chlorophyll-a concentration at Ibitinga reservoir along the 2005 hydrologic year and (2) assess how changes in hydraulic residence time (HRT) affect the spatial distribution of phytoplankton blooms at Ibitinga Reservoir. The study site was chosen because previous studies reported seasonal changes in the reservoir limnology which might be related to the reservoir seasonality and hydrodynamics. Six Landsat/TM images were acquired over Ibitinga reservoir during 2005 and water flow measurements provided by the Brazilian Electric System National Operator - ONS were used to compute the reservoir´s residence time, which varied from 5.37 to 52.39 days during 2005. The HRT in the date of image acquisition was then compared to the distribution of chlorophyll-a in the reservoir. The results showed that the HRT increasing implies the increasing of the reservoir surface occupied by phytoplankton blooms.


Resumo As imagens de satélite são frequentemente usadas para a identificação de florações de fitoplâncton porque sua presença causa mudanças significativas na cor da água. A abundância das florações pode ser quantificada por medidas de concentração de clorofila-a. Diversos estudos sugerem o uso da razão de reflectância das bandas TM4/TM3 Landsat, para determinar as concentrações de clorofila-a em sistemas aquáticos. Este trabalho tem como objetivos: (1) aplicar um modelo semi-empírico que usa essa razão para mapear a concentração de clorofila-a no reservatório de Ibitinga ao longo do ano hidrológico de 2005; (2) avaliar como as mudanças no tempo de residência hidráulica afetaram a distribuição de florações na superfície do reservatório. O reservatório de Ibitinga foi selecionado porque estudos prévios indicavam mudanças sazonais nas propriedades limnológicas do reservatório, as quais poderiam estar relacionadas à sazonalidade e à hidrodinâmica. Seis imagens TM/Landsat foram adquiridas sobre o reservatório de Ibitinga durante o ano de 2005. Foi então usada uma tabela associando intervalos de razão de banda a intervalos de concentração de clorofila-a. Medidas de vazão fornecidas pelo Operador Nacional do Sistema Elétrico – ONS foram utilizadas para calcular o tempo de residência hidráulica do reservatório, que variou entre 5,37 e 52,39 dias durante 2005. O tempo de residência hidráulica em cada data de aquisição da imagem foi então comparado com a área ocupada pelas florações de fitoplancton. Os resultados indicaram uma forte relação entre o tempo de residência hidráulica e a área ocupada por florações. Em junho de 2005, quando o reservatório atingiu seu menor tempo de residência hidráulica, apenas 20% de sua área estiveram ocupadas por florações. Em setembro e outubro, quando a residência hidráulica atinge o seu máximo, mais que 80% da superfície do reservatório foram ocupadas por florações de fitoplancton.

BACKGROUND: Cadmium (Cd) is well known as one of the most toxic metals affecting the environment and can severely restrict plant growth and development. In this study, Cd toxicities were studied in strawberry cv. Camarosa using pot experiment. Chlorophyll and malondialdehyde (MDA) contents, catalase (CAT), superoxide dismutase (SOD), ascorbate peroxidase (APX) activities and mineral nutrient concentrations were investigated in both roots and leaves of strawberry plant after exposure Cd. RESULTS: Cd content in both roots and leaves was increased with the application of increasing concentrations of Cd. We found higher Cd concentration in roots rather than in leaves. Chlorophyll a and b was decreased in leaves but MDA significantly increased under increased Cd concentration treatments in both roots and leaves. SOD and CAT activities was also increased with the increase Cd concentrations. K, Mn and Mg concentrations were found higher in leaves than roots under Cd stress. In general, increased Cd treatments increased K, Mg, Fe, Ca, Cu and Zn concentration in both roots and leaves. Excessive Cd treatments reduced chlorophyll contents, increased antioxidant enzyme activities and changes in plant nutrition concentrations in both roots and leaves. CONCLUSION: The results presented in this work suggested that Cd treatments have negative effect on chlorophyll content and nearly decreased 30% of plant growth in strawberry. Strawberry roots accumulated higher Cd than leaves. We found that MDA and antioxidant enzyme (CAT, SOD and APX) contents may have considered a good indicator in determining Cd tolerance in strawberry plant.

ABSTRACT

The current macroscopical and microscopical signs in the fresh drowning victim are non-specific. Moreover, putrefaction will vanish these autopsy findings quite rapidly. The study of additional detecting methods to diagnose drowning has become one of the hottest topics in forensic medicine. This article reviewed several methods commonly used in the determination of drowning in forensic practice. The advantages and disadvantages of various methods were also discussed.

ABSTRACT

The rosewood (Aniba rosaeodora Ducke) is a native tree species of Amazon rainforest growing naturally in acidic forest soils with reduced redox potential. However, this species can also been found growing in forest gaps containing oxide soils. Variations in the forms of mineral nitrogen (NO3- or NH4+) may be predicted in these different edaphic conditions. Considering that possibility, an experiment was carried out to analyze the effects of different NO3-:NH4+ ratios on the growth performance, mineral composition, chloroplastid pigment contents, photochemical efficiency photosystem II (PSII), and nitrate redutase activity (RN, E.C.1.6.6.1) on A. rosaeodora seedlings. Nine-month-old seedlings were grown in pots with a washed sand capacity of 7.5 kg and submitted to different NO3-:NH4+ ratios (T1 = 0:100 percent, T2 = 25:75 percent, T3 = 50:50 percent, T4 = 75:25 percent, and T5 = 100:0 percent). The lowest relative growth rate was observed when the NO3-:NH4+ ratio was equal to 0:100 percent. In general, high concentrations of NO3- rather than NH4+ favored a greater nutrient accumulation in different parts of the plant. For the chloroplastid pigment, the highest Chl a, Chl b, Chl tot, Chl a/b and Chl tot/Cx+c contents were found in the treatment with 75:25 percent of NO3-:NH4+, and for Chl b and Cx+c it was observed no difference. In addition, there was a higher photochemical efficiency of PSII (Fv/Fm) when high NO3- concentrations were used. A linear and positive response for the nitrate reductase activity was recorded when the nitrate content increased on the culture substrate. Our results suggest that A. rosaeodora seedlings have a better growth performance when the NO3- concentrations in the culture substrate were higher than the NH4+ concentrations.


O pau-rosa (Aniba rosaeodora Ducke) habita, naturalmente, solos florestais ácidos com potencial redox reduzido. No entanto, estas espécies têm sido encontradas também em clareiras que, teoricamente, apresentam solos mais oxidados. Nestas diferentes condições edáficas, pode-se prever variações nas formas do nitrogênio mineral (NO3- ou NH4+). Considerando essa possibilidade foi conduzido experimento com o objetivo de estudar os efeitos de diferentes relações de NO3-:NH4+ sobre o acúmulo de biomassa, a composição mineral, as concentrações de pigmentos cloroplastídicos, a eficiência fotoquímica do fotossistema II e a atividade da enzima redutase do nitrato (RN, E.C.1.6.6.1) em A. rosaeodora. Plantas jovens com nove meses foram cultivadas em vasos com capacidade para 7,5 kg e submetidas a diferentes relações de NO3-:NH4+ (T1 = 0:100 por cento, T2 = 25:75 por cento, T3 = 50:50 por cento, T4 = 75:25 por cento e T5 = 100:0 por cento). As menores taxas de acúmulo de biomassa foram observadas quando a relação das concentrações de NO3-:NH4+ foi igual a 0:100 por cento. De maneira geral, a maior concentração de NO3- em relação a NH4+ favoreceu maior acúmulo de nutrientes nas diferentes partes da planta. Para os teores de pigmentos cloroplastídicos, os maiores valores de Chl a, Chl tot, Chl a/b e Chl tot/x+c, foram encontrados no tratamento com 75:25 por cento de NO3-:NH4+, não ocorrendo diferença para os teores de Chl b e Cx+c. Em relação a eficiência fotoquímica do fotossistema II (Fv/Fm) maior eficiência também ocorreu quando predominou maior concentração de NO3- em relação ao NH4+. Quanto a atividade da reductase do nitrato observou-se resposta linear e positiva para o incremento da concentração de nitrato no substrato de cultivo. Portanto, os resultados obtidos sugerem que as plantas jovens de A. rosaeodora apresentaram melhor desempenho quando as concentrações de NO3- no substrato de cultivo foram superiores as concentrações de NH4+.
